After weight measurements and counting, eight fish from each tank were sampled, individually anesthetized with MS-222 (20 mg/L) (tricaine methanesulfonate, Western Chemicals, Inc., Ferndale,Washington) and sacrificed for tissue samples collection for biochemical assays, nutrients body composition and digestibility measurements.Intestines, livers and skins from O. niloticus were excised and subsequently homogenized in a glass beds with 9 volumes (v/w) of 0.8% physiological saline. The homogenate was centrifuged (2500 rpm, at 4 °C for 10 min), the resulting supernatant was collected and subsequently frozen at −80 °C for biochemical assays according to manufacturers' instructions. One whole intestine, liver and a piece of skin from each fish were used in all analyses. On the days of analyses,malondialdehyde (MDA) concentration was determined by using enzyme-linked immune sorbent assay (ELISA) kit (Hengyuan, Shanghai).Protease, lipase, amylase, lactate dehydrogenase (LDH), glutathione Stransferase (GST), superoxide dismutase (SOD), total antioxidant capacity (TAC), acid phosphatase (ACP), and alkaline phosphatase (AKP)were measured by using specific commercial assay kits (Nanjing Jiancheng Institute, China), with their results recorded on a microplate reader (Epoch, BioTek, USA). The remaining fish (with viscera removed) were used for body composition and apparent nutrients digestibility measurements according to standard methods (AOAC, 1995).
